BioCryst-Lab (Bari)
The laboratory is equipped with advanced instrumentation for the crystallization of proteins (both soluble and membrane proteins) and for the characterization of biological samples and nanomaterials for structural analysis.
It is equipped with an Oryx4 crystallization robot (Douglas Instruments, UK), featuring an LCP (Lipidic Cubic Phase) module. The instrument supports several crystallization techniques, including vapor diffusion (sitting-drop and hanging-drop), microbatch screening, optimization, and microseeding.
The system provides extensive screening capabilities, allowing the evaluation of a wide range of experimental conditions. It is possible to modulate the pH (from 3 to 9), use different precipitating agents such as PEGs, polymers, and salts, and operate within a temperature range from 4°C to 30°C.
In collaboration with the Bari branch of the Institute for Sustainable Plant Protection (IPSP) of the National Research Council of Italy (CNR), the laboratory has also been equipped with a SpectroLight 600 DLS system (Xtal), capable of determining the hydrodynamic radius (Rh from 0.5 nm to 1 μm), the polydispersity index (PDI), and diffusion coefficients (D). Measurements are performed using multi-angle acquisitions (50–173°) based on signal autocorrelation and are complemented by an optical imaging system for continuous monitoring of crystal growth.
The laboratory also hosts an extensive library of commercial crystallization screens for identifying optimal crystallization conditions, including specialized kits for soluble proteins, membrane proteins, and macromolecular complexes, listed below:
