Cell Biology

The cell biology laboratory is equipped with all instruments necessary for mammalian cell cultures, primary or continuous lines, both of tumor origin and not. The behaviour of single cells as well as cell populations is explored by the use of numerous technical approaches. These include: immunocitological tecniques analyzed by the means of intra/extracellular flow citometry and epifluorescence microscopy; biochemical immunoassay for the detection of signalling events; viability assay; detection of cellular dynamics and and traffiking by live cells imaging at single cells level, detection of apoptotic marks, oxidative stress often in conjunction with the use of viability dyes either by flow cytometry or epifluorescence microscopy or live cell imaging and analysis.

Class II laminar flow cabinet

A class II laminar flow cabinet (Termo fisher safe 2020), a cabin in which the air is filtered by HEPA (High Efficiency Particulate Air) filters, and which allows you to manipulate biological material in sterility, reducing the possibility of crossed contamination, (sample operator and vice versa).

CO2 incubator

CO2 incubator, (Heracell), which allows to obtain an atmosphere suitable for in vitro cultures of cells and tissues. In addition to maintaining a temperature of 37 ° C, the cabin dispenses and controls CO2 at the optimal concentration to allow the maintenance and growth of mammalian cells.

Centrifuge (Allegra Backmann)

Centrifuge (Allegra Backmann) allows to separate and wash the cells from the culture medium at all stages of maintenance and experimentation.

Inverted fluorescence microscope

Inverted fluorescence microscope Leica DMI6000 B is equipped with standard optics, (10x, 20x, 40x, and 63x underwater), with a digital camera and with a combination of DIC (Differential Interference Contrast) and internal modulation (IMC) systems, that allows to combine contrast techniques in transmitted light with fluorescence, to carry out observations on cultured live cells and on traditional preparations. It is also furnished with a whole series of advanced hardware and software controls of the fire, the stage, the outflow filters (Ultra Fast Internal Filter Wheel (IFW). It also has a FIM (Fluorescence Intensity Manager) system that allows the regulation of the ” fluorescence light intensity and limits the area of the sample affected by the fluorescence avoiding prolonged irradiation, thus reducing photobleaching.

Cytofluorimeter (Partec CyFlow® ML)

Cytofluorimeter (Partec CyFlow® ML) is a multiparametric analyzer of cells suspension that allows to analyze two different forward scatter signals and one side scatter (giving information on the shape, size and roughness of the cells) and combining them with 13 different fluorescence channels. Data acquisition and analysis is managed through dedicated FloMax® software for Windows. The CyFlow® ML is equipped with a special system that presses the absolute volumetric counts. Different systems based on diodes, solid state lasers or Nd: YAG lasers are used in combination to obtain a wide range of excitation wavelengths.

Incucyte® SX1 (Sartorius)

An automated system for acquiring and analyzing images from a cell or tissue culture in physiological environment; it operates from inside a CO2 incubator. The instrument integrates phase contrast with fluorescence allowing to carry out both standard protocols for the control of cellular functionality (cell counts, cytotoxicity, proliferation, apoptosis, migration, etc.) and advanced investigations for applications in various fields (immunohistochemistry, neuronal activity, angiogenesis, tumor spheroids, phagocytosis, internalization of antibodies, etc). IncuCyte is equipped with 4X, 10X, 20X obiectives; three software modules include: cell by cell analysis, enables label-free cell count and cell-by-cell classification; Chemiotaxis analysis, enables analysis of label-free and fluorescently labeled chemotactic cell migration; NeuroTrack Analysis, enables analysis of neurite dynamics with or without labels.
