Sviluppo di una piattaforma tecnologica per l’identificazione di modulatori allosterici di S1R con potente attività anti sars-cov-2
PB28, a known sigma-1 (s1R) antagonist, showed significant antiviral activity against SARS-CoV-2 (IC90 = 0.28 μM). After testing the PB28 analogues in our hands, a direct correlation between s1R affinity and SARS-CoV-2 antiviral activity was not observed suggesting the presence of an allosteric mechanism occurring only if specific ligand-protein interactions take place. We aim at shedding light on such mechanism to rationally design new s1R ligands with potent SARS-CoV-2 antiviral activity.
